The CRCM's flow cytometry platform, located at the Institut Paoli-Calmettes, provides the scientific community with access to advanced flow cytometry technologies, including both conventional and spectral systems, ranging from high-content analysis to single-cell sorting. The platform is open to internal CRCM teams as well as external academic and industrial partners.

Since 2015, the platform has adhered to a rigorous quality management system and is certified ISO 9001:2015 and NFX 50-900:2016, ensuring the use of high standard for the production of reliable and reproducible data.

Its core missions include both technical and scientific support by training users in instrument operation, assisting with the development of complex high content panels, and supporting data analysis. The platform supports researchers throughout every stage of their flow cytometry projects.

In close collaboration with the Immunomonitoring Platform (PIC), it contributes to the development of innovative fundamental and translational research projects, thereby enhancing the impact of cross-sectional studies conducted at the Institut Paoli-Calmettes.

Available equipment: 3 flow cytometers, 3 cell sorters, and a Bio-Plex 200 (Bio-Rad).

  • Scientific Supervision: Provided by Daniel Olive, with the support of a steering committee comprising research team representatives, an administrative representative, and the center director or their delegate.

Platform equipment

Sorters
Two sorting cytometers under the control of platform personnel:
  • An Aria III SORP(Becton Dickinson: 5 lasers 350/405/488/561/640 nm - 20 PMT) in a fume hood.
  • An Aurora CS(Cytek Biosciences: 5 lasers 350/405/488/561/640 nm - 64 APD) in a fume hood.

A self-service sorting cytometer:
  • A Melody(Beckton Dickinson): 3 lasers 405/488/561 - 10 PMT) in a hood.

Analyzers
3 self-service cytometer analyzers :
  • A Fortessa(Becton Dickinson: 3 lasers 405/488/630 nm - 14 PMT) with automated plate reader.
  • Symphony A3(Becton Dickinson: 5 lasers 350/405/488/561/630 nm - 28 PMT) with automated plate reader.
  • Aurora spectral technology(Cytek Biosciences: 5 lasers 355/405/488/532/630 nm - 64 APD) with automated reader for plates and tubes.
This equipment is accompanied by services and technological development activities. The technical platform plays an essential role in the training, collection and dissemination of knowledge in flow cytometry.
Software
  • 1 PC equipped with FlowJo and Spectroflo software.
  • 1 PC equipped with KALUZA, FlowJo and Diva software.
  • 1 OMIQ license.
Bioplex 200

This technology, which combines the principles ofELISA and flow cytometry, enables the simultaneous assay of numerous protein or nucleic targets in small sample volumes.
