CRCM\"\/><\/a><\/figure>\n\n<h2 class=\"wp-block-heading\">Antonia Boudet<\/h2>\n\n<h4 class=\"wp-block-heading\">Doctoral student<\/h4>\n\n<h2 class=\"wp-block-heading\">Our laboratory studies the role and targeting of oncogenic protein kinases, and identifies new vulnerabilities in hematological diseases.<\/h2>\n\n<p><strong>Hematopoiesis<\/strong> is a model of differentiation that enables us to<strong>study the molecular mechanisms<\/strong> involved in the transition from a highly regulated system, in terms of both<strong>homeostasis and differentiation<\/strong>, to a deregulated system leading to proliferative syndromes or <strong>leukemias.<\/strong> <br\/>In this context, we are particularly interested in<strong>identifying the mechanisms<\/strong>, pathways and protein players initiated by transforming mutations in protein kinases such as <strong>KIT<\/strong> and <strong>FLT3<\/strong>, and their impact in various pathologies linked to this receptor (hemopathies, mastocytosis, gastrointestinal stromal tumors &#8211; GIST, melanomas). We are also studying other original kinases of interest in oncology: <strong>CDK6, FES and FER.<\/strong> <\/p>\n\n<p>Our project is based on <strong>three<\/strong> main <strong>approaches<\/strong> in which we have recognized expertise:<\/p>\n\n<ul class=\"wp-block-list\">\n<li><strong>Functional identification<\/strong> of activating mutations in receptors. This expertise, acquired with <strong>KIT in mastocytosis<\/strong>, forms the basis for collaborative projects with clinical research teams. We also search for mutations affecting other therapeutic targets, in particular <strong>kinases<\/strong>, in other <strong>tumor pathologies<\/strong>.  <\/li>\n\n\n\n<li>The search for the involvement of new specific substrates\/pathways linked to <strong>oncogenic signaling,<\/strong> downstream of activated Receptor <strong>Tyrosine Kinases<\/strong>, using proteomics or <strong>siRNA<\/strong> approaches. Our expertise in gene transfer in <strong>hematopoietic progenitors is a major asset<\/strong> in this functional approach.<\/li>\n<\/ul>\n\n<p>Contribution to the identification and characterization of <strong>tyrosine kinase<\/strong> inhibitors, in collaboration with AB Science. We are involved in the optimization of <strong>kinase-selective<\/strong> compounds <strong>of interest in targeted therapy.<\/strong> Our ambition is to identify <strong>new therapeutic pathways or targets<\/strong>, by combining our expertise and collaborations. <\/p>\n\n<p>Our work covers both <strong>basic<\/strong> and <strong>translational<\/strong> <strong>research<\/strong> <strong>.<\/strong> To this end, we have access to <strong>numerous<\/strong> human and mouse cell <strong>models<\/strong>, 3D in vitro cell models, mouse models of hematological diseases, and access
