DNA damage is a major cause of the point mutations that lead to cancer. <\/h2>\n\n<p><strong>Mechanisms of lesion tolerance :<\/strong><\/p>\n\n<p>Despite efficient repair systems, the replication fork often encounters residual lesions. Cells have two strategies for tolerating lesions: <\/p>\n\n<ul class=\"wp-block-list\">\n<li>translational lesion synthesis (TLS), whereby a specialized DNA polymerase inserts a few nucleotides opposite the lesion, with the risk of introducing mutations;<\/li>\n\n\n\n<li>the Damage Avoidance (DA) pathway, which is non-mutagenic since it relies on mechanisms related to homologous recombination with the sister chromatid.<\/li>\n<\/ul>\n\n<p>The balance between these two pathways is very important, as it defines the level of mutagenesis during replication of a damaged genome. We have developed a new technique enabling us to insert a single lesion at a specific site on the bacterial chromosome<em>(Escherichia coli)<\/em>. This approach enables us to quantitatively monitor <em>in vivo<\/em> the distribution between translesion synthesis (TLS) and damage bypass (DA) in different genetic backgrounds, in order to determine the genes that regulate the balance between these two pathways. We are also analyzing the early replication intermediates that form at the lesion site, using molecular approaches (qPCR, 2D agarose gel, electron microscopy), to define the mechanisms involved in lesion tolerance pathways.   <\/p>\n\n<p>We have developed a similar approach in eukaryotic yeast cells<em>(Saccharomyces cerevisiae<\/em>).
