To preserve genomic integrity, cells have mechanisms for detecting and repairing DNA damage. These mechanisms play a crucial role, since unrepaired DNA damage can lead to mutations, chromosomal rearrangements or loss of genetic material, all of which promote tumorigenesis.  <\/p>\n\n<p><strong>Structure-specific DNA endonucleases (SSEs) are key to maintaining genome integrity.<\/strong>  These enzymes are essential molecular scissors for the management of DNA secondary structures formed during DNA replication, DNA repair or recombination processes, and transcription.<\/p>\n\n<p>However, any reaction involving a cut in the DNA carries risks, opening the door to the formation of chromosomal rearrangements at the origin of the carcinogenesis process. It is therefore essential that the action of SSEs is closely coordinated with downstream mechanisms to ensure that the original chromosome structure is fully restored. It is also vital for genome stability that ESS action is rigorously channeled to avoid anarchic action.  <\/p>\n\n<p><strong>Our team has been studying the spatio-temporal regulation of SSEs for many years.<\/strong> Using the yeast <em>Schizosaccharomyces pombe<\/em> as a model organism, we have identified several pathways controlling the Mus81-Eme1 endonuclease in response to DNA damage.<\/p>\n\n<p><strong>At the same time, our team was a pioneer in the identification of the human SLX4 protein, the study of which now constitutes the core of our activity.<\/strong>  We have demonstrated that SLX4 acts as a platform for the coordination and control of the XPF-ERCC1, MUS81-EME1 and SLX1 nucleases. SLX4 plays a key role in the coordination of many other factors important for genome maintenance and cell cycle control, with which it also interacts<em> (Figure 1)<\/em>. <\/p>\n\n<figure class=\"wp-block-image\"><img decoding=\"async\" src=\"https:\/\/crcm-marseille.fr\/wp-content\/uploads\/2025\/01\/figure_1_multifunctional_scaffold_equipe_controle-des-endonucleses_crcm.png\" alt=\"\"\/><\/figure>\n\n<p><strong><em>Figure 1:  <\/em><\/strong><em>SLX4 is a multifunctional scaffold that fulfils key functions in genome maintenance by interacting with and coordinating the action of a number of important DNA damage response, DNA repair, and cell-cycle control factors.
